The ordered sequence will be assembled by overlapping synthetic oligonucleotides and amplfied by PCR. The longer the sequence the higher the oligonucleotide quality requirements needed to obtain the correct clone.
The PCR product is cloned into a suitable screening vector using standard restriction sites. Most screening vectors of our manufactures are pUC or pBluescript based. In all cases the final vector will be an ampicilin or kanamycin resistant high copy plasmid.
Clones are screened by sequencing. After a 100% correct clone is identified, a plasmid preparation of a single colony is performed. The DNA will be re-sequenced to ensure correctness.
